    Rather than a nice flat area we have one with many large ‘clumps’ of grass. Will it be necessary to remove these by hand or just cover with tarp etc to kill off for an extended period

    Those clumps will decline and rot in the absence of light, though may leave lumps of soil that has built within them, so you can level that probably more easily after the grass is dead.
    Certainly no need to remove them by hand.
